生物化学与分子生物学系


学术研究

 

当今,生命科学已经进入到分子生物学时代。医学作为生命科学的主要分支,它首先面对着两个基本问题:(1)对健康的理解和维护;(2)对疾病的有效诊断和治疗。在分子生物学时代,疾病发生的分子机制,疾病的预防,疾病的诊断和治疗都依赖于生物化学与分子生物学的理论和技术。随着各学科的相互交叉与渗透,本学科在几十年的发展历程中,逐步形成了稳定突出的学科特色和科研方向。

(1) 肿瘤发生的分子机制和干预防治:早在20世纪50-70年代,本学科的老一辈教授在营养与癌(食管癌的早期诊断、病因学研究)、自由基与肿瘤等研究方面取得了颇多成就。1978年老前辈研究小组与美国新泽西医学院杨中枢教授协作开展了对林县食管癌的研究,并承担的国家八五科技攻关项目食管癌的早期诊断其中分课题研究,于1996年获国家计委、国家科委和国家财政部联合颁发的国家八五科技攻关重大科技成果奖。① 食管癌的病因学研究(安玉会教授)在国家六五和七五重大攻关课题食管癌的病因学研究中,从林县食管癌高发区高污染霉菌中分离出了交链胞霉毒素,为霉菌毒素致食管癌的理论奠定了分子病因学基础。在随后的研究中,安玉会教授先后开展了芦笋,大豆黄酮,灵芝三萜和姜素干预肿瘤干细胞增殖和侵润的一系列化学预防研究。其研制的芦笋饮料和口服液获国家发明专利并开发生产。② 骨肿瘤的分子生物学发病机理及基因调控和干预防治(李月白教授等):多年以来,对人的骨肉瘤、骨巨细胞瘤等骨肿瘤的基因表达调控及防治方面进行有价值的深入系列研究。通过对骨肉瘤、骨软骨瘤和正常骨组织中的抑癌基因p21wafl /cip1基因表达,P27cyclinE ki-67蛋白等表达检测分析研究,揭示了它们在不同组织中的表达差异与骨肉瘤的发生、发展及临床预后的相关性。通过对骨肉瘤细胞干预研究,揭示了通过激活PPAR信号通路可抑制骨肉瘤细胞的生长、增殖并诱导其分化和凋亡。为恶性骨肿瘤的发生机制及防治途径提供了实验依据,其研究获得科技厅科研资助,科研成果获得科技进步成果奖及发表多篇研究论文。

(2) 氧毒性相关疾病的分子机制和干预防治及植物化学物生物学效应研究(王明臣教授等):此研究系统揭示了煤矽肺发生的氧毒性分子机制及干预防治路径,研究成果获得河南省医药卫生科技成果一等奖。与加拿大农业部合作,对万寿菊中的植物化学物叶黄素(lutein)进行了生物学效应的综合系统研究,揭示了叶黄素干预阻断恶性肿瘤、动脉硬化的发生及发展和视力促进作用的分子机制,研究成果被国际食品化学权威杂志 Food chem. Toxiol 刊发。

(3) 心血管疾病的分子生物学发病机理与防治研究(臧明玺教授等):前辈曾对动脉粥样硬化发生机制以及中药的治疗进行研究,揭示高脂可导致动脉粥样硬化的发生并发现多种中药对此疾病有疗效,获河南省教育厅科技成果二等奖。对高血压病及其脑并发症防治的研究,获河南省科技进步奖。目前,臧明玺教授对心脏发育、分化及疾病的调控机制进行了深入研究,并获得了重要进展。本研究获得国家863课题及国家自然科学基金等多项研究课题资助,发表了多篇SCI文章,并获得多项科技进步奖励。

(4) 骨疾病的分子生物学发病机理和干预防治研究(李月白教授等)多年以来,对疾病的发生机理与防治进行了系列研究。对股骨头坏死及骨质疏松症的发病机理、基因表达调控及防治进行了深入研究。揭示RNA干扰技术或miRNA靶向基因调控成脂关键基因PPARγ,高效阻止细胞成脂分化,促进其成骨分化,为股骨头坏死的发生机理提供了有价值的理论依据、早期防治的新靶点和策略。本研究获得3项国家自然科学基金项目、1项卫生部科研基金项目、4项省级研究项目资助,分别获得河南省科技进步二等奖、三等奖。

本学科将继续坚持业已形成的学科特色和科研方向,将不断在上述研究中取得新进展。学科中拥有河南省学术技术带头人1人、河南省教育厅学术技术带头人2人。近年来承担国家自然基金课题8项、863计划研究项目2项、教育部和人事部留学基金3项、河南省课题12项等, 相关研究成果已经发表在国际主流期刊 ,如Cell Mol Life SciRNA BiolJ Mol BiolFood chem toxicol等。在郑州大学实施一省一校及基础医学院加强学科建设的过程中,本学科均被列入重点建设内容。目前,随着高水平基础设施和分子生物学及细胞生物学技术平台的构建,本学科在肿瘤防治及心肌再生等领域的研究将独具特色。
