学术研究

一、研究方向

1、临床药理:主要进行药物代谢与药物动力学、CYP2E1与肿瘤等炎症相关疾病研究等,目的是为临床个体化用药、以及肿瘤等炎症相关疾病防治提供依据。在药物代谢与药物动力学研究方面,创建了国内领先、国际先进的肝脏CYP450药物代谢的体外研究技术平台,对CYP450的生理和病理进行了系统研究,证明CYP450基因多态性以及酶含量对药物代谢影响的有限性,为指导临床个体化用药奠定了基础;肝癌患者肝CYP450各亚型代谢活性改变不一,有的降低,有的不变,有的增高;发现CYP2E1为炎症反应中的新靶点,证明其与肿瘤等炎症相关性疾病的发生发展有关,并研发了CYP2E1特异性抑制剂。上述研究得到了重大新药创制国家科技重大专项和5项国家自然科学基金项目的资助,所创制的国家1.1类抗脑缺血新药正在进行临床研究,相关研究成果被编入人民卫生出版社出版的研究生教材《高级药理学》中

2、遗传药理:该方向依托河南省高校药物基因组学重点实验室,在药物代谢、代谢调控及药物基因组学等方面已有近20年的研究积累;团队已建立完善的药物基因组学、代谢组学、表观遗传学检测和分析平台,重点围绕肝脏药物代谢酶的个体差异和表达调控,结合“药物基因组学”和“表观遗传学”的原理,从遗传多态性、表观遗传调控及发育早期环境因素等方面开展一系列研究,以期揭示个体对药物不同反应的机制、药物代谢酶基因表达调控机制等。近年来,团队致力于揭示表观遗传调控因子如miRNAs、组蛋白修饰和lncRNAs等在药物代谢酶个体差异性表达、发育表达及外源物诱导表达中的调控机制,取得了原创性的研究结果,形成了明显的优势。基于药物代谢酶的表达具有“时空性”,团队专注于核受体/代谢酶/转运体介导下的内/外源物代谢调控对疾病发生发展转归、合理用药及发育早期药物暴露对药物代谢酶表达的影响,对指导临床精准用药具有重要理论意义。以上相关研究获得了科技部国际合作项目、国家重点研发计划子课题、国家自然科学基金、河南省杰出人才等科研项目的资助。

3、肿瘤药理:主要进行外泌体与肿瘤标志物、消化道肿瘤及微管蛋白聚合抑制剂等方面的研究。在外泌体与肿瘤标志物研究方面,依托郑州大学“外泌体与肿瘤标志物研究中心平台”,采用多种方法联合应用筛选人体液外泌体中的肿瘤标志物,分别在细胞水平、动物模型及临床水平验证筛选出来的肿瘤标志物应用于液体活检的可行性;在消化道肿瘤研究方面,近年来通过体外和体内试验,从分子、基因、蛋白和细胞生物学行为等多个方面对其发病机制、侵袭转移及逆转肿瘤多药耐药进行了深入研究,为寻找消化道肿瘤转移和评价预后的分子标志物以及发现消化道肿瘤潜在的分子治疗靶点提供新思路;同时,还进行了微管蛋白聚合抑制剂、NEDDylation抑制剂的设计、合成、活性评价等研究,近年来设计合成得到相关500余个并探讨了其中10余个重点化合物的作用机制,并首次报道了NEDDylation激动剂作为抗胃癌药物的机制

4、神经药理:我们的神经药理研究包括慢性疼痛发生机制的研究、小胶质细胞在神经发育和疾病中的作用研究以及新药药理研究。在慢性疼痛发生机制的研究方面,我们参与制作了国际上第二个癌痛小鼠模型,并对其发生机制进行多方面的探索。在小胶质细胞在神经发育和疾病中的作用研究方面,我们对星型胶质细胞和小胶质细胞在大鼠神经痛发生机制中的作用进行了初步探讨,同时研究小胶质细胞中P2X受体以及其相互作用蛋白在神经疾病发生过程中的作用。在新药药理研究,近年来通过体内和体外研究,开发并申请了国家1·1类新药:新型抗脑缺血药物6--3-正丁基-13H-异苯并呋喃酮的研发。不仅深入研究开发治疗脑缺血新药,同时通过新药药理学的研究寻找脑缺血的发病机制,为临床合理用药提供实验及理论依据。

5心血管药理:主要方向为心血管相关疾病的机制研究,研究领域包括:心脏方面,探讨心肌缺血性疾病的发病机制,通过建立心肌梗死、缺血再灌注和基因敲除鼠的动物模型,从免疫反应参与心肌缺血损伤的角度,探讨NLRP3炎性小体活化参与心肌重构的分子机制,同时从离子通道和缝隙连接蛋白角度研究心肌缺血后心律失常发生的病理机制,从而为心肌缺血性疾病寻求新的药物治疗靶点。血管方面,通过应用组织特异性表达的模式动物,开展在血管内皮和平滑肌细胞中表达的前列腺素E2受体4亚型(EP4)在血管重塑、高血压以及高血压肾病中的作用及相关机制的研究。已获得国家自然科学基金面上项目一项,青年基金两项,河南省自然科学基金一项。

6、药物设计及纳米药物:1)计算驱动的创新药物发现与作用机制研究:通过计算生物学、计算化学、生物信息学、分子生物学、药物化学、结构生物学等多学科交叉手段,从源头开展创新药物发现。发现药物靶标的构象动态变化机制、新口袋和新分子,并在分子、细胞和动物水平开展分子作用机理研究,为疾病的诊断和治疗提供新的探针分子、新的药物作用位点和新的先导结构,为开发具有自主知识产权的创新药物奠定基础。近年来,系统研究了G蛋白偶联受体的动态构象和激活机制,发现一批具有开发前景的活性化合物,研究结果在NatureScienceNature Structural & Molecular BiologyNature CommunicationsJournal of Medicinal ChemistryJournal of Biological Chemistry等国际权威杂志发表。(2)结合纳米技术、生物化学、纳米生物学和肿瘤药理学方面的理论知识和技术手段,研究纳米载体的设计、构建以及其在肿瘤药理研究中的作用。(3)纳米酶和铁蛋白的生物医学应用,探索铁蛋白药物载体和纳米材料酶学特性在肿瘤诊疗上的应用。
