The macromolecular ribonucleoprotein, spliceosome catalyzes nuclear pre-mRNA splicing and confers complexity of proteins. My recent work has uncovered essential RNA structural features in the spliceosomal RNA core for catalysis, and important RNA structural dynamics during catalytic activation. These findings deepen the understanding of structure-function relations of ribonucleoprotein and renew the knowledge of recent cryo-EM structures of spliceosome. Also I found that novel function of the RNA helicase of Prp2, acts as an RNPase during the catalytic activation stage, and is unexpectedly negatively regulated by a protein complex of RES during early assembly stage. This ensures efficient splicing and proofreads spliceosome for active conformation. The findings unfold the fundamental mechanisms for the production of mRNA and prevent genome-scale aberrant proteins, which underlies a growing number of human diseases from developmental disorders to cancer.
包鹏辉博士,1980年1月出生于河南漯河,2002年武汉大学生命科学学院生物技术专业学士毕业,2009年取得武汉大学生物化学及分子生物学专业博士学位。随后在德国马克斯普朗克生物物理化学研究所的著名生物学家Reinhard Lührmann实验室从事生物大分子-spliceosme的研究,先后在《Genes & Development》《Nucleic Acids Research》等国际顶尖刊物发表论文。
